MECHANISM OF ACTION

Long R3 IGF-1 analog: substitution of Arg at position 3 plus a 13-amino acid N-terminal extension. It minimizes affinity for IGF-binding proteins (IGFBP), drastically prolonging the half-life to 20–30 h compared to 12–15 min for native IGF-1. Full activation of the IGF-1 receptor: tyrosine kinase → MAPK, PI3K/Akt → protein synthesis, cell proliferation, and glucose uptake.
