MECHANISM OF ACTION

Universal cellular cofactor in its oxidized form. It accepts electrons in glycolysis and the TCA cycle, forming NADH, and fuels oxidative phosphorylation. It serves as a substrate for sirtuin deacetylases (SIRT1–7), which regulate longevity; PARP, DNA repair enzymes; and CD38/cADPR, involved in calcium signaling. Its levels decrease by 40–50% by middle age.

SIDE EFFECTS AND SAFETY

IV: flushing, headache, and nausea if infused too quickly; it should be administered by drip over 2–4 hours. SubQ: discomfort at the injection site. Oral forms are largely ineffective; oral precursors such as NMN/NR are preferred.
